More information would be extremely helpful. Like this is on your S5, how many miles, has any work been done recently, and do you have any shifter upgrades?
I would start by looking at the shifter and then the shift linkage where it connects to the transmission. First check that the hardware is tight and then check for play. The factory shifter base and shift linkage use plastic balls to allow for movement that can wear out causing play like what you described.
